An Aptamer Glue Enables\nHyperefficient Targeted Membrane\nProtein Degradation

Targeted membrane\nprotein degradation (TMPD) offers significant\ntherapeutic potential by enabling the removal of harmful membrane-anchored\nproteins and facilitating detailed studies of complex biological pathways.\nHowever, existing TMPD methodologies face challenges such as complex\nmolecular architectures, scarce availability, and cumbersome construction\nrequirements. To address these issues, this study presents a highly\nefficient TMPD system (TMPDS) that integrates an optimized bivalent\naptamer glue with a potent protein transport shuttle. Utilizing this\napproach, we successfully degraded both the highly expressed protein\ntyrosine kinase 7 in CCRF-CEM cells and the poorly expressed PTK7\nin MV-411 cells. This system represents significant advancement in\nthe field of molecular medicine, offering a new avenue for targeted\ntherapeutic interventions and the exploration of cellular mechanisms.
