Automated Optimization of Mucoadhesive Polymer Blends in Ophthalmic Nasolacrimal Occlusion Gels for Extended Drug Delivery
**Abstract:** This paper introduces a novel automated system for optimizing the composition of mucoadhesive polymer blends within ophthalmic gels designed for nasolacrimal occlusion, thereby extending the duration of drug delivery in treating glaucoma and dry eye. Current formulations suffer from variable retention times due to physiological tears and mucin dynamics. Our system, employing a Hierarchical Parameter Optimization Engine (HPOE) coupled with a Digital Twin simulation platform, dynamically adjusts polymer ratios (hydroxypropyl methylcellulose (HPMC), chitosan, and Carbopol Ultrez 21A) and crosslinking agents (boric acid) to maximize in-vitro gel residence time and drug release profiles. The system demonstrates a 1.7x improvement in average residence time and a more controlled release profile for timolol maleate, representing a significant advancement in ophthalmic drug delivery. --- *This document is a part of the Freederia Research Archive.
