FORMULATION AND EVALUATION OF PULSATILE DRUG DELIVERY SYSTEM OF SIMVASTATIN

In the present research work, we have designed a pulsatile formulation of Simvastatin to treat dyslipidemia pattern of the disease. Core tablets were prepared by incorporating different concentration of disintegrants and were compressed in between different concentration of polymers. The core and compression coated tablets were subjected to pre-formulation, physicochemical and In-vitro drug release studies. Core formulations, characterized by different release rates and mechanisms, will be coated by compression with an outer shell of different polymeric barrier layers of polymers. The coatings prevent drug release from the core until the polymeric shell is completely eroded or swollen. FTIR studies revealed that there was not any chemical reaction between pure drug Simvastatin and polymers. The pre and post-compressional parameters of tablets were also found to be within limits. Our optimized formulation F-3 releases Simvastatin 100.15 % up to 12 hours. Key words: Pulsatile formulation, Simvastatin, Sodium starch glycolate, Chitosan, Eudragit RSPO, HPMC K 100 and Compression coated tablets.
