Development of A Novel Floating Tablet of Lafutidine for Sustained Release in Phosphate Buffer [Ph – 6.8]
This study presents the development of a novel floating tablet formulation of Lafutidine designed forsustained drug release in phosphate buffer (pH 6.8). Targeting enhanced gastric retention and prolongedtherapeutic effect, the formulation utilizes a gas-generating mechanism with sodium bicarbonate and ahydrophilic polymer matrix, primarily HPMC, to ensure buoyancy and controlled release. Tablets wereprepared via direct compression and evaluated for floating behavior, drug release kinetics, andphysicochemical characteristics. The optimized formulation exhibited a floating lag time of less than oneminute and sustained drug release over 12 hours, aligning with a non-Fickian diffusion model. Thisinnovative approach not only addresses Lafutidine’s short half-life and site-specific absorption but alsooffers a promising strategy for improving its bioavailability and therapeutic efficacy in managing acidrelated disorders .
