Combination Strategy\nof Two Hydrogels Loaded with\nPrussian Blue Nanoparticles and Calcium Peroxide Separately for Diabetic\nWound Repair

Oxidative stress and excessive hypoxia are two key factors\nhindering\ndiabetic wound healing. Prussian blue nanoparticles (PBNPs) and calcium\nperoxide (CPO) possess the characteristics of scavenging reactive\noxygen species (ROS) and chemically generating oxygen respectively,\nand both can be used as biomedical materials. However, when the two\nsubstances coexist in the aqueous media, Prussian blue undergoes decomposition.\nIn this study, gelatin/carboxymethyl chitosan @PBNP hydrogel and gelatin/carboxymethyl\nchitosan@CPO hydrogel were prepared separately and combined into concentric\ncircular structures. The results showed that the combination of two\nhydrogels has good properties for swelling, adhesion, in vitro antioxidant,\nscavenging intracellular ROS, downregulating tumor necrosis factor-α\n(TNF-α), and enhancing the proliferation and migration of fibroblasts.\nIn a diabetic mouse wound model, the combined application group shortened\nwound healing time, promoted growth of granulation tissues, and accelerated\ncollagen formation compared with a single hydrogel treatment. The\ncombined use of the two drug-loaded hydrogels has potential application\nin promoting diabetic wound repair.
