Dynamic Microvesicle Engineering via Feedback-Controlled Lipid Raft Modulation for Enhanced Gene Delivery

**Abstract:** This paper introduces a novel approach to enhance gene delivery efficiency via dynamic modulation of microvesicle (MV) composition and targeting capabilities. Current MV-based gene delivery systems often display inconsistent efficacy due to heterogeneity in MV size, lipid composition, and target cell interaction. We propose a feedback-controlled system leveraging microfluidic devices and real-time optical imaging to manipulate lipid raft formation within donor cells, thereby influencing MV biogenesis and subsequent gene transfer. This approach allows for dynamic tuning of MV properties to optimize targeting and intracellular delivery. The system promises a 10x increase in transfection efficiency compared to traditional MV-based methods with improved safety profiles and adaptability to diverse cell types and gene payloads.
