Magnetic Nanocarriers\nfor pH/GSH/NIR Triple-Responsive\nDrug Release and Synergistic Therapy in Tumor Cells

In this study, the\nmesoporous Fe3O4 nanodrug\ncarriers containing disulfide bonds (CHO-SMNPs) were successfully\nsynthesized and characterized. Doxorubicin (DOX) was loaded onto the\nCHO-SMNPs as a model drug and gatekeeper through the formation of\nimine bonds with the aldehyde groups on the surface of the mesoporous\nmaterials. This drug carrier demonstrates effective drug release triggered\nby pH, glutathione (GSH), and near-infrared (NIR) light, along with\nsatisfactory photothermal conversion efficiency under NIR irradiation\nat 808 nm. Furthermore, CHO-SMNPs exhibit excellent blood compatibility\nand biodegradability. They also show good biocompatibility and efficient\ncellular uptake in HeLa and MCF-7 cancer cells. Most importantly,\nthe CHO-SMNPs/DOX has shown significant effectiveness in killing both\nHeLa and MCF-7 cancer cells. Consequently, CHO-SMNPs/DOX presents\nsubstantial potential as a magnetic-targeted, pH/GSH/NIR triple-triggered\ndrug delivery system for synergistic chemo-photothermal therapy in\ntumor treatment.
