A Collection\nof Novel Antitumor Agents That Regulate\nLipid Metabolism in the Tumor Microenvironment
Lipid metabolism disorder is the cause of one of the\nmost significant\nmetabolic changes in tumors. In the process of tumor occurrence and\ndevelopment, tumor cells choose a continuous metabolic adaptation\nto accommodate the changing environment to the maximum extent possible.\nIn a variety of tumors, the uptake, production, and storage of lipids\nare generally upregulated. Tumor cells take advantage of lipid metabolism\nto access basic energy, biofilm components, and signal molecules of\nthe tumor microenvironment required for proliferation, survival, invasion,\nand metastasis. This Perspective briefly uncovers the main metabolic\nprocesses and key factors involved in lipid metabolism reprogramming,\nmainly related to lipid uptake, <i>de novo</i> synthesis\nand storage of fatty acids, oxidation of fatty acids, cholesterol\nsynthesis, and related regulatory factors. From a medicinal chemistry\nperspective, agents against related key targets are reviewed, expecting\nto pave the way for promising antitumor drugs with prospects for application\nthrough lipid metabolism reprogramming.
