Compressible\nHollow Microlasers in Organoids for High-Throughput\nand Real-Time Mechanical Screening
Mechanical\nstress within organoids is a pivotal indicator in disease\nmodeling and pharmacokinetics, yet current tools lack the ability\nto rapidly and dynamically screen these mechanics. Here, we introduce\nbiocompatible and compressible hollow microlasers that realize all-optical\nassessment of cellular stress within organoids. The laser spectroscopy\nyields identification of cellular deformation at the nanometer scale,\ncorresponding to tens of pascals stress sensitivity. The compressibility\nenables the investigation of the isotropic component, which is the\nfundamental mechanics of multicellular models. By integrating with\na microwell array, we demonstrate the high-throughput screening of\nmechanical cues in tumoroids, establishing a platform for mechano-responsive\ndrug screening. Furthermore, we showcase the monitoring and mapping\nof dynamic contractile stress within human embryonic stem cell-derived\ncardiac organoids, revealing the internal mechanical inhomogeneity\nwithin a single organoid. This method eliminates time-consuming scanning\nand sample damage, providing insights into organoid mechanobiology.
