Cyclic Enzymatic\nSignal Amplification-Driven DNA Logic\nNanodevices on Framework Nucleic Acid for Highly Sensitive Electrochemiluminescence\nDetection of Dual Myocardial miRNAs
MicroRNAs (miRNAs) have emerged as promising biomarkers\nfor acute\nmyocardial infarction (AMI). There is an urgent imperative to develop\nanalytical methodologies capable of intelligently discerning multiple\ncirculating miRNAs. Here, we present a dual miRNA detection platform\nfor AMI using DNA logic gates coupled with an electrochemiluminescence\n(ECL) response. The platform integrates DNA truncated square pyramids\nas capture probes on gold-deposited electrodes, enabling precise quantification\nof miRNA associated with AMI. The cyclic enzymatic signal amplification\nprinciple of strand displacement amplification enhances the miRNA\ndetection sensitivity. AND and OR logic gates have been successfully\nconstructed, enabling intelligent identification of miRNAs in AMI.\nCalibration curves show strong linear correlations between ECL intensity\nand target miRNA concentration (10 fM to 10 nM), with excellent stability\nin consecutive measurements. When applied to clinical serum samples,\nthe biosensor exhibits consistent performance, underscoring its reliability\nfor clinical diagnostics. This innovative approach not only demonstrates\nDNA nanotechnology’s potential in biosensing but also offers\na promising solution for improving AMI diagnosis and prognosis through\nprecise miRNA biomarker detection.
