Effective Nose-to-Brain\nDelivery of Blood-Brain Barrier\nImpermeant Anti-IL-1β Antibody via the Minimally Invasive Nasal\nDepot (MIND) Technique

Treatment of neuroinflammation and neurodegenerative\ndiseases using\nbiologic therapies is limited due to the blood-brain barrier (BBB).\nThis study explores a clinically validated approach to bypass the\nBBB for the purposes of direct central nervous system (CNS) delivery\nof antibodies using the Minimally Invasive Nasal Depot (MIND) technique.\nUsing a lipopolysaccharide (LPS)-induced mouse model of neuroinflammation,\nwe evaluated the efficacy of MIND in delivering a BBB impermeant full-length\nanti-IL-1β antibody. The results demonstrated that MIND delivery\nresulted in a significant reduction in IL-1β levels and microglial\nactivation in relevant brain regions, notably outperforming conventional\nintravenous (IV) administration. These results underscore the ability\nof the MIND approach to transform the treatment landscape for a range\nof neurodegenerative diseases by enabling the targeted delivery of\notherwise BBB impermeant therapeutics.
