A NOVEL XYLANASE ENZYME PRODUCTION FROM COCOS NUCIFERA USING SOLID STATE FERMENTATION

Xylanase is a naturally occurring enzyme begins in microbes and fungi. Xylanase belonging to pentosanases, a class of enzyme which breaks cell wall matrix of plants breaks down xylan into xylose. Xylanases also find its application in various industries, include textile, paper industry, beverage industry, bakery industry, pharmaceuticals industry, and biofuel produce. The main aim of the present research work was optimization of process parameters for the production of Xylanase enzyme using cocos nucifera leaves as substrate in a solid-state fermentation by using microorganism Trichoderma harzianum. For the production of Xylanase enzyme different parameters like incubation time, incubation temperature, pH, inoculum level, and moisture content and were optimized. The incubation time of 72hrs, the temperature of 250C, pH 7, inoculum level of 0.8%v/w, and moisture content of 0.1%v/w, were marked optimum for the production of Xylanase. Different carbon components were screened for the enzyme production; they are glucose, fructose, lactose, maltose and sucrose used as carbon supplements. Among the carbon source, glucose gave good production when compared with other carbon supplements. To determine the nitrogen effect on enzyme production, potassium nitrate was taken and 0.1%w/w was observed optimum for the enzyme production. Final conclusion was that cocos nucifera could be a promising substrate for industrial application since it producea significant Xylanase (86.84IU/ml) activity in solid state fermentation.
