Recent Advancements in Naphthofuran Derivatives as Anti-Cancer Agents
Cancer remains a leading cause of mortality worldwide, underscoring the urgent need for novel and effective therapeutic strategies. Naphthofurans, a class of heterocyclic compounds possessing a fused furan and naphthalene ring system, have emerged as promising candidates for anticancer drug development due to their diverse biological activities and structural versatility. This review provides an overview of recent advancements in the design, synthesis, and evaluation of naphthofuran derivatives as anticancer agents. We will examine the various mechanisms of action targeted by these compounds, including inhibition of enzymes, modulation of signaling pathways, induction of apoptosis, and disruption of the tumor microenvironment. Furthermore, we will highlight recent structure-activity relationship (SAR) studies, preclinical efficacy, and potential strategies for optimizing naphthofuran-based anticancer therapies.
