CRISPR/Cas9 based HLA reduction of T cells for universal T cell therapy
Adoptive cell therapy (ACT) with CAR-T cells is successful in cancer/infection treatment. Due to the high cost and complexity of production, gene-modified allogeneic cell sources are proposed. This work demonstrates that HLA knock-out cells retain their effector function but are prone to rejection and cannot present antigens. Selective HLA editing offers multi-donor compatibility and overcomes the aforementioned limitations. HLA-edited T cells are shown here to be also effective in vivo.
