Ni-Catalyzed\nAsymmetric Reductive Arylation of α‑Substituted\nImides

α-Aryl imides\nare common structural motifs in bioactive molecules\nand proteolysis-targeting chimeras designed for targeted protein degradation.\nAn asymmetric Ni-catalyzed reductive cross-coupling of imide electrophiles\nand (hetero)aryl halides has been developed to synthesize enantioenriched\nα-arylglutarimides from simple starting materials. Judicious\nselection of electrophile pairs allows for coupling of both electron-rich\nand electron-deficient (hetero)aryl halides in good yields and enantioselectivities.

Abstract

α-Aryl imides\nare common structural motifs in bioactive molecules\nand proteolysis-targeting chimeras designed for targeted protein degradation.\nAn asymmetric Ni-catalyzed reductive cross-coupling of imide electrophiles\nand (hetero)aryl halides has been developed to synthesize enantioenriched\nα-arylglutarimides from simple starting materials. Judicious\nselection of electrophile pairs allows for coupling of both electron-rich\nand electron-deficient (hetero)aryl halides in good yields and enantioselectivities.
