Integrating\nVascular Phenotypic and Proteomic Analysis\nin an Open Microfluidic Platform
This research introduces\na vascular phenotypic and proteomic analysis\n(VPT) platform designed to perform high-throughput experiments on\nvascular development. The VPT platform utilizes an open-channel configuration\nthat facilitates angiogenesis by precise alignment of endothelial\ncells, allowing for a 3D morphological examination and protein analysis.\nWe study the effects of antiangiogenic agentsbevacizumab,\nramucirumab, cabozantinib, regorafenib, wortmannin, chloroquine, and\npaclitaxelon cytoskeletal integrity and angiogenic sprouting,\nobserving an approximately 50% reduction in sprouting at higher drug\nconcentrations. Precise LC-MS/MS analyses reveal global protein expression\nchanges in response to four of these drugs, providing insights into\nthe signaling pathways related to the cell cycle, cytoskeleton, cellular\nsenescence, and angiogenesis. Our findings emphasize the intricate\nrelationship between cytoskeletal alterations and angiogenic responses,\nunderlining the significance of integrating morphological and proteomic\ndata for a comprehensive understanding of angiogenesis. The VPT platform\nnot only advances our understanding of drug impacts on vascular biology\nbut also offers a versatile tool for analyzing proteome and morphological\nfeatures across various models beyond blood vessels.
