Exploring the bioactive potential of 3-hydroxy-3-phenyl-isoindolin-1-one: A DFT-based computational study

The compound, 3-hydroxy-3-phenyl-isoindolin-1-one (3HPIO): was studied to explore its structural, electronic, spectroscopic, thermodynamic, and bioactive properties. A comprehensive computational method employing the Density Functional Theory (DFT) approach in the gaseous phase was adopted to drive the most stable optimized geometry. The RMSD scores of 0.993 and 0.983 confirmed the structural similarities between theoretical and experimental datasets. The molecule’s reactive sites were analyzed through Molecular Electrostatic Potential (MEP) and Electron Localization Function (ELF) analysis. Frontier Molecular Orbital (FMO) analysis defined orbitals with the highest and lowest occupied energy. The energy gap between HOMO and LUMO was found to be 5.37 eV. The UV-vis studies under gas and other solvent mediums provided the required energy for the maximum absorption of wavelengths through electronic transitions. Non-linear optical (NLO) properties explored the 3HPIO’s optical properties, making it optically active. RDG and Hirshfeld surfaces along with 2D finger-plots showed the compound’s molecular interactions. In the process of docking, the 4HBM protein which belongs to the ligase domain was found to be highly competent with an observed binding energy of -7.7 kcal/mol. The molecular dynamic simulations provided a more refined insight into the solubility, stability, and interactions. The compound 3HPIO was observed to be a bioactive molecule and is a viable option for future research and development, as depicted in the analyses made through drug-likeness
