Development and Optimization of Bilastine-Loaded Lipid Polymer Hybrid Nanoparticles for improved Solubility using Central Composite Design
ABSTRACT: In order to enhance the solubility of bilastine (BL) for the treatment of allergic rhinitis, Lipid Polymer Hybrid Nanoparticles (LPHNs) were created and evaluated. Nanoprecipitation approach created these adaptable Lipid Polymer Hybrid Nanoparticles from Bilastine (BL), oleic acid, Soluplus and Tween 60. Surface charge, particle size, and entrapment efficiency (EE%) all showed LPHNs. Ex vivo permeation, stability, zeta potential, Fourier transform infrared, and in vitro drug release studies were used to describe the LPHNs. The optimized BL- LPHNs had a zeta potential of -47.7 mV and a polydispersity index of 0.293. Entrapment efficiency for optimized BL-LPHNs was 94%. The F-12 formulation has two times the ex vivo BL permeability of the pure drug. Compared to commercial formulations and pure drugs, optimized BL-LPHNs have demonstrated greater drug release over a 24-hour period. Based on physicochemical properties and in vitro characterizations, the LPHNs enhanced BL solubility.
