An Adaptive, Bio-Integrated Primer Editing System Utilizing Microfluidic Control and Real-Time Error Correction (A-BI-PEC)
**Abstract:** The limitations of current prime editing (PE) technologies, specifically off-target effects and reliance on exogenous guide RNAs, hinder broad clinical application. A-BI-PEC introduces a novel, self-correcting PE system integrating microfluidic control for precise reagent delivery, bio-integrated guide RNA expression, and a real-time error correction circuit based on CRISPR interference (CRISPRi). This system significantly reduces off-target events, improves efficiency, and demonstrates potential for autonomous genome editing in complex biological environments. Our approach represents a direct commercialization path towards safer and more effective genome editing therapies, projecting a $5 billion market within 5 years, particularly in treating inherited genetic disorders.
