Assessment of Oxidative Stress of Ethanoic Extract of Ripe and unripe Banana fruits in salt loaded Alcoholic and indomethacin induced Ulcerative Rats

An alternative disease treatment approach is the research of medicaments from traditional medicine. Plant with antioxidant capabilities are used as herbal treatments for ulcer diseases. This study aims to examined the Oxidative Stress of Ethanoic Extract of Ripe and unripe Banana fruits in salt loaded, Alcoholic and indomethacin induced Ulcerative Rats. Eighty male wistar rats (weighing between 120-140g were grouped into 16 standard study groups of 5 animals each. Group A: Control group this group received food and water only. Group Bi: Ulcer induced by indometacine (NSAID) without treatment Group Bii: Ulcer (Alcohol) without treatment Group C: Received Sodium chloride orally for 2 weeks without treatment, Group Di: Received Sodium chloride orally all through the study (NaCl)+ Ulcer (NSAID) without treatment Group Dii: Received Sodium chloride orally all through the study (NaCl)+ Ulcer (Alcohol ) without treatment, each of the group were treated with unripe banana extract and ripe banana extract, all animals were sacrificed after exposure to chloroform. Blood was collected through cardiac puncture, placed in a lithium heparin sample bottles. Blood samples was spun in a bucket centrifuge at 2500 RPM (rounds per minute) for 10 minutes after which plasma was collected and stored frozen in plain sample bottles and was analyzed for Oxidative stress Biomarkers (GSH, GST, SOD, CAT and MDA) levels by spectrophotometric method. Data obtained were analyzed as mean ± standard deviation using one way analysis of variance (ANOVA. Level of significance between groups was taken as p<0.05. It was observed in the present study significant increase in GPX in the test group NaCl+RIPED BANANA+ULCER (NSAID) when compared to the ULCER (NSAID) group and Nacl group respectively. This is suggesting that ripped bananas could be involved in detoxification. The present study have demonstrated the antioxidant activity of both ripe and unripe banana. There was significant decrease in GSH and CAT levels in the group that was loaded with NaCl before Ulcer induction using NSAID (NaCl+ULCER (NSAID) when compared to the group that was loaded with NaCl before Ulcer induction using Alcohol (NaCl+ULCER (ALCOHOL) this is an indication that NSAID might be more toxic to gastric tissues than alcohol. There was no significant difference in GSH, GPX, CAT and MDA levels when (NaCl+UNRIPED BANANA) group was compared to (NaCl+RIPED BANANA), this suggests that both ripped and unripe banana exhibits the same effect on oxidative stress of cells. it was also observed that ulceration induced by alcohol might activates events that leads to reduction of CAT.
