DNA barcoding of bacteria with Sanger Sequencing
This module integrated different topics to proceed with DNA barcoding using Sanger sequencing: 1) Concept of DNA barcoding, 2) Degenerated primers, 3) Cellular lysis, 4) PCR, 5) Pipetting, 5 & 6) Electrophoresis, 7) Sanger sequencing, 8) DNA subway (Mosquito data), 9) DNA subway (Bee data), 10) Oral Presentation of the DNA barcoding students will have done. It is sometimes difficult for students to visualize the position of primers on the primary and complementary DNA strand, as well as the direction of each primer on the strand (3' to 5' vs. 5' to 3.). To tackle this issue, the student has the option here to decide between two concepts of exercises (meteorite & aliens OR Indigenous' feeding resources, but it can be done with other types of concepts). They have to "barcode" manually a DNA sequence with a set of primers that is given to them, but first, they need to understand the position, the direction, and the complementarity between primers and DNA strands.
