Abstract 6870: Rhenium complex-induced cytotoxicity in triple negative breast cancer cells (TNBC)
Abstract Triple negative breast cancer (TNBC) remains the most aggressive subtype with major obstacles to effective management necessitating continuous search for new pharmacological agents. This includes resistance to therapeutic agents including carboplatin. We hypothesize that novel Rhenium reagents will overcome resistance to carboplatin in TNBC cells. We investigated the cytotoxic potentials of synthesized rhenium complexes, PR6 and PR7, on a panel of TNBC cell lines (BT-549, MDA-MB-231, HCC 1806, MDA-MB-468), including carboplatin-resistant HCC1806. Cells were treated with 2.5µM PR6 and 1.5µM PR7 for 72 hours, and MTS-based cell viability assay was performed. The expression levels of selected metastasis promoting proteins including HMGA2 and phospho-AKT (p-AKT) in cell lysates were determined at baseline and post-treatment using western blot analysis. Our results indicate that PR7 is more potent than PR6 in significantly reducing cell viability. Also, all TNBC cells investigated are sensitive to PR7 and PR6 treatment except for parental HCC 1806 and carboplatin-resistant HCC 1806 that are resistant to PR6 treatment. We also observe higher expression of HMGA2 and p-AKT proteins in carboplatin-resistant HCC 1806 cells as compared to parental cells. In conclusion, PR7 rhenium compound may be a promising agent for TNBC management and overcoming carboplatin resistance.
