Therapeutic Effect of Moringa Oleifera Leaves Extract Against Hepatotoxicity in Diabetic Rat

Background: Streptozotocin (STZ) is a cytotoxin specific to beta cells in the pancreas that directly damages organs and results in diabetes. The liver is among the many organs that can be harmed by STZ-induced organ toxicity. The current study studied the effects of extract from Moringa Oleifera leaves on rats' liver damage caused by streptozotocin. The goal of this study was to examine whether or not male albino rats given extract from Moringa oleifera leaves may avoid contracting diabetes mellitus owing to streptozotocin (STZ). Materials and Methods : Seven groups of 42 male albino rats were randomly assigned. Group I rats were left untreated,Group II rats were given oral glibenclamide (5 mg/kgb.wt./day), while Rats in Group III were given oral MOLE at a dose of 200 mg/kg b.wt./day. One intraperitoneal injection of STZ (45 mg/kg diluted in saline solution) was given to the rats in group IV (the diabetic group). Group V: Oral MOLE (200 mg/kg b.wt./day) was given to diabetic rats. Group VI: Oral glibenclamide (5 mg/kg + 200 mg/kg body weight/day) was given to diabetic rats. Group VII: The same dosage of glibenclamide and MOLE were given orally to diabetic rats. All of the animals were sacrificed after 45 days. Results: Results showed remarkable increases in liver function (AST and ALT) activities, kidney function (urea and creatinine) levels, TNFα and IL-1β levels and liver MDA level, in rats with diabetes versus rats without the disease. Additionally, findings revealed that diabetic rats exhibited a considerable decrease in total protein and antioxidants markers (GSH, SOD CAT) activities, in contrast to the group under authority. Oral delivery of MOLE, Glibenclamide and their combination to diabetic untreated rats led to remarkable enhancement on all mentioned parameters. The diabetic group had vacuolations, hazy swelling, hydropic degeneration, and signet-ring fatty alterations, among other hepatocellular degenerative changes of varying degrees, according to histopathological analysis. Groups treated with MOLE showed notable improvement because of MOLE had a therapeutic effect on liver dysfunction and oxidative stress induced by diabetes, probably because of its anti-inflammatory and antioxidant properties. Conclusion: It is possible that MOLE could help prevent the metabolic abnormalities brought on by diabetes due to its antioxidant properties.
