Ultrastructural changes in eggshell during incubation: mechanisms and implications

The avian eggshell is a complex, multi-layered bioceramic structure that performs mechanical, physiological, and biochemical functions essential for embryo survival. Its design represents a fine balance between protection and permeability, ensuring successful incubation and hatching. Ultrastructural changes in the eggshell during incubation are primarily due to controlled demineralization and microstructural reorganization mediated by the chorioallantoic membrane. These changes facilitate calcium mobilization, gas exchange, and embryonic development, with direct implications for hatchability, shell strength, and chick viability. Incubation leads to a progressive decline in eggshell quality in terms of thickness, hardness, and mineral content. However, these changes are physiologically advantageous, they optimize the balance between mechanical protection early in incubation and increased permeability and hatchability toward the end. The transformation of the eggshell reflects a highly regulated adaptation that ensures successful embryonic development and hatching. One of the most significant changes during incubation is the progressive resorption of calcium from the inner shell layers, particularly the mammillary layer, to support skeletal formation in the growing embryo. This process alters the ultrastructure of the shell, affecting parameters such as thickness, porosity, crystal orientation, and mechanical integrity. These changes can have important implications for hatchability, shell strength at the time of hatching, and post-hatch chick quality.
