Expanding spectral multiplexing panels with ease using new StarBright Dyes with novel spectral signatures 3515
Abstract Description Bio-Rad’s 32 StarBrightTM Dyes for flow cytometry have superior brightness and low spillover, with the ability to be able to be fixed and used in any buffer with no loss in signal. These dyes can be premixed and stored for long periods of time prior to use with no changes to the staining. With full spectrum flow cytometry increasing panel sizes there is a need for high-quality fluorophores, that will provide flexibility when building panels, improving panel design and data. Here we describe four novel StarBright Dyes, excitable by the ultraviolet 355 nm or violet 405 nm lasers; StarBright Violet 535, StarBright UltraViolet 540, StarBright UltraViolet 700 and StarBright UltraViolet 770. They have unique spectral signatures tailored for spectral instruments to fill gaps in dye emission wavelengths currently available. Data generated using a Cytek Aurora (Cytek Biosciences) and an ID7000 (Sony Biotechnology) cytometer highlight the properties and performance in single stains and a multiplexing panel. These new dyes are compatible with both the current range of 32 StarBright Dyes and other commonly used fluorescent dyes in flow cytometry. Having all the benefits of StarBright Dyes, they are ideal for panel expansion and give increased choice when building panels of any size.
