Automating Antibody Cocktail Preparation and Sample Staining with the C-FREE Pluto LT system 9074
Abstract Description The preparation of antibody cocktails for flow cytometry, especially for intracellular staining, is time-consuming and prone to variability. The C-FREE Pluto system offers an automated solution to streamline this process, reducing human error, minimizing reagent waste, and enhancing reproducibility. The Pluto LT workstation automates staining, washing, and master-mix preparation, providing a comprehensive “all-in-one” sample preparation solution. At the Stanford Immune Monitoring Core, we successfully evaluated the Pluto LT system for surface and intracellular staining assays, confirming its reliability and reproducibility. Building on this, we are exploring the system’s potential for automating antibody master-mixing with a complex multicolour panel, focusing on feasibility, stability, and time-saving benefits. This investigation supports our broader efforts to advance high-dimensional flow cytometry, ensuring the rigor required for accurate intracellular staining and rare cell population analysis. Our findings show that the Pluto LT system reduces preparation time and variability compared to manual methods, while ensuring consistent stability across multiple antibody cocktails. The automation of this process has also increased assay efficiency, allowing technicians to focus on downstream analysis. This implementation aligns with our commitment to advancing immune monitoring by enhancing both reliability and efficiency in flow cytometry applications.
