Impact of DSS-induced chronic colitis on emphysema via IL-1ß pathways in mice 4690
Abstract Description Studies have suggested a link between ulcerative colitis and pulmonary dysfunction. However, comprehensive studies exploring the relationship between colitis and COPD remain limited. Male C57BL/6J mice were used for production of DSS-induced chronic colitis or cigarette smoke (CS)-induced emphysema model. During experimental period, the body weight of mice was measured along with observation of stool characteristics. the MLI score in the DSS group was similar to that of the CS-exposed emphysema group. It has been suggested that even if mild, chronic colitis can cause pulmonary emphysema. In mesenteric lymph nodes, CD11b and F4/80 positive populations were significantly more abundant in DSS groups compared to other groups. In lung tissue, CD49b or βTCR/CD3 expressing cells were more detected in colitis or emphysema group compared to control group while CD4 or CD25/CD3 positive cells was significantly less in colitis groups compared to control group. In addition, it found that IL-1β expression in BALF, lung and colon was higher in DSS-induced colitis group compared to control group. Treatment with anti-IL1β antibody (αIL-1β) lowered MLI values in DSS + αIL-1β group were lower compared to both DSS and DSS + IgG group, indicating that the inhibition of IL-1β function might improve DSS-induced colitis. In conclusion, this study demonstrated that DSS-induced colitis significantly impacts the pulmonary emphysema throughout the activity of IL-1β.
