Resveratrol enhances anti-tumor function of Cisplatin and Rapamycin when treating cancer B cell lines 9388

Abstract Description Resveratrol is a plant metabolite with proven anti-cancer properties in B cell malignancies without damaging normal cells. Unfortunately, the anti-tumor properties of resveratrol are modest. Standard treatments against cancer, such as cisplatin are effective but toxic to normal cells. The goal of this study was to explore the effect of resveratrol in combination with low dosages of anti-cancer compounds with the objective of managing B cell malignancies. To achieve this goal, we tested two B cell lines: Nor20, an EBV transformed B cell line and SUP-B15, an acute lymphoblastic leukemia cell line and examined the viability of these cells in response to resveratrol (100 µM), cisplatin (1 µM), rapamycin (25 µM), and combinations of resveratrol/cisplatin or resveratrol/rapamycin. Our key observations show that Nor20 and SUP-B15 viability dropped with resveratrol (89%, 81.5%), cisplatin (82.2%, 43.2%), rapamycin (92.7, 92.9%), resveratrol/cisplatin (68.4%, 35.2%) and resveratrol/rapamycin (80.2%, 80.9%), respectively, when normalized to their controls. In addition, using flow cytometry we observed that Nor20, at these concentrations, CD19 expression did not change significantly but SUP-B15 cells decreased expression of CD19 with cisplatin treatment (70.9%) and the resveratrol/cisplatin (79.9%) co-treatment. We conclude that although these compounds combined with resveratrol or alone can decrease viability of both B cell malignant cell lines, the mechanism of action differs.
