Single-cell full-length TCR/BCR and CITE-Seq multiomic analysis with pre-designed lyophilized T and B cell panels enable comprehensive T and B cell characterization 4490

Abstract Description Studying B and T cell heterogeneity can be enhanced by using highly multiplexed DNA-barcoded antibodies with full-length TCR/BCR assay for simultaneous profiling of cell-surface proteome, transcriptome and TCR/BCR clonal information from single cells. This approach increases insights into T and B-cell receptor functionality and role in diverse immune cell populations. While high-plex panels are powerful tools for profiling complex cell populations, the barrier for adoption is high due to labor- and cost-intensive validation and optimization. To address this, 30-plex lyophilized T cell and B cell panels were designed at optimal concentrations to analyze cell type specific immune response conveniently and flexibly. The 30-plex T and B lyophilized panels were combined and used with the BD Rhapsody™ TCR/BCR Next Multiomic assay to profile sample-tagged resting and stimulated PBMCs. Results show consistent pairing efficiency performance when compared to the 60-plex liquid cocktail. The multiomic data show reliable detection of distinct T and B subpopulations with paired receptor sequences and gene expression profiles. This demonstrates the compatibility of the pre-designed panels with the BD Rhapsody™ TCR/BCR Next Multiomic assay to analyze clonotype-specific phenotypes. With fewer manual steps and standardized CITE-Seq staining, these panels simplify workflows and enable consistent results, critical for scaling up to higher cell and sample numbers. Abstract
