Overcoming Challenges in AAV Plasmid Preparation: Advanced Solutions for Superior ITR Integrity and High-Yield Production 9427
Abstract Description AAV-based gene therapies are transforming treatments for genetic disorders, cancer, and rare diseases. However, preparing high-quality AAV plasmids is challenging, particularly in maintaining ITR integrity due to its repetitive, high GC-content nature. Regulatory agencies require intact ITRs for clinical applications, as mutations can compromise vector performance. To address this, GenScript developed the GenStable™ ITR strain and AAV applied vectors, ensuring >95% ITR integrity through 10 generations—a 100% improvement over conventional methods—while boosting plasmid yields fourfold. To address this, GenScript developed the GenStable™ ITR strain and AAV applied vectors, ensuring >95% ITR integrity through 10 generations—a 100% improvement over conventional methods—while boosting plasmid yields fourfold. By integrating optimized plasmid solutions and advanced QC technologies, GenScript streamlines AAV workflows, reduces costs, and accelerates high-quality AAV vector production, driving innovation in gene therapy development.
