Using proteomic surrogates to improve understanding of cardiometabolic risks in IMIDs 3653

Abstract Description Patients with immune mediated inflammatory diseases (IMIDs) have an increased risk for cardiovascular (CV) disease and metabolic disorders, which are often underpredicted by traditional risk calculators. These analyses aimed to determine whether previously validated proteomic surrogates of CV event risk, kidney health risk, dementia risk, and other key cardiometabolic (CM) measures can provide a better understanding of CM risks associated with IMIDs. SomaSignal® tests (derived from ∼5000 plasma protein measurements using the SomaScan® assay) were applied to a subset (n = 1,743) of samples from the IMID-Biobank at VHIR, including healthy control participants (n = 254) and patients with ulcerative colitis (UC; n = 280), Crohn’s disease (CD; n = 294), rheumatoid arthritis (RA; n = 304), psoriatic arthritis (PsA; n = 304), and plaque psoriasis (PS; n = 307). An exploratory analysis using ANOVA between patients with an IMID, and healthy controls was conducted using a raw p-value cutoff of < 0.05. CV and dementia risk were significantly elevated in UC, CD, RA, and PS compared to healthy controls and risk correlated with disease severity in most cases. Kidney health risk was significantly worse in UC, CD, and RA compared to healthy controls. Multiple CM measures including VO2max, glucose tolerance and visceral fat significantly worsened in IMID patients compared to healthy controls. Together these results suggest SomaSignal® tests may provide a useful tool for assessing CM risk in IMID patients. Topic Categories Translational and Interventional Immunology (TI)
