Overcoming challenges in mRNA plasmid preparation: advanced solutions for Poly(A) tail integrity and high-yield production 9428

Abstract Description The rapid growth of mRNA research is driving breakthroughs in vaccine development, cancer immunotherapy, and regenerative medicine, increasing the demand for high-quality plasmid DNA templates. These templates are crucial for in-vitro transcription (IVT), yet propagating Poly(A)-containing plasmids in E. coli remains a major challenge. Researchers face low plasmid yields, Poly(A) tail instability, and prolonged development timelines, hindering therapeutic progress and increasing costs. The Poly(A) tail is essential for mRNA stability, translation efficiency, and downstream applications. To address these challenges, GenScript has developed proprietary mRNA vectors with prebuilt Poly(A) tail sequences, ensuring optimized templates for IVT. Paired with the GenStable™ Poly(A) strain, specifically engineered for Poly(A) plasmid propagation, these solutions achieve >90% success in generating intact Poly(A) sequences through 8 generations, representing a 300% improvement over conventional methods and increasing plasmid yields by over 7-fold. These cutting-edge plasmid solutions enhance mRNA production efficiency, reduce costs, and accelerate therapeutic research, enabling faster, more reliable mRNA-based advancements.
