Optimization of a T, ILCs, MAIT and NK cells multicolor panel with CytoFLEX mosaic 63 spectral detection module 9426
Abstract Description This study presents the optimization of a spectral flow cytometry panel designed to comprehensively characterize T cells, Innate Lymphoid Cells (ILCs), Mucosal-Associated Invariant T (MAIT) cells, and Natural Killer (NK) cells using the new CytoFLEX mosaic 63 spectral detection module, which provides spectral capabilities for our CytoFLEX S flow cytometer. The panel was designed to include markers that define each of these key immune cell populations, enabling simultaneous identification of multiple subsets with minimal spectral overlap. By leveraging the unique advantages of spectral flow cytometry, which allows for the detection of a higher number of parameters in a single sample, this panel provides a detailed, multi-dimensional analysis of immune cell differentiation, activation, and function. The optimization involved selecting appropriate fluorochromes, designing optimal antibody combinations, and ensuring precise unmixing. The enhanced resolution and sensitivity of the spectral flow cytometer enabled the detection of rare or subtle immune subsets that are difficult to identify with conventional flow cytometry techniques. This optimized panel offers significant advantages for studying T cells, ILCs, MAIT cells, and NK cells in diseases. The integration of spectral flow cytometry in immunological studies paves the way for more detailed and accurate immune profiling, ultimately contributing to a better understanding of immune regulation and therapeutic development.
