Powerful end-to-end spatial analyses using precise spatial proteomics 4179

Abstract Description True spatial biology supports our understanding of the progression and treatment of disease by distilling complex cellular interactions into actionable spatial signatures. We leveraged the new enhanced photobleaching in cyclic immunofluorescence (EpicIF) technology for CellScape™ Precise Spatial Proteomics to develop a pipeline pairing the platform with fluorescent biomarker panels targeting key structural features and phenotypes to enable accurate downstream data analysis for immunology and immuno-oncology applications. The panels were compatible with the EpicIF workflow, which allows the use of commercially available antibody-fluorophore conjugates already deployed in other techniques. Our data demonstrate the functionality of Segmentation, Immune Profiling, Tissue Architecture, and Myeloid Cell Profiling assay panels using a range of human FFPE tissues. The Segmentation panel clearly defines cell membrane boundaries for precise segmentation of most cell types. The Immune Profiling panel facilitates the characterization of immune infiltrates and the determination of both composition and proximity to tissues of interest. The Tissue Architecture panel targets markers identifying tissue subtypes and layers, and the Myeloid panel facilitates tumor characterization by profiling myeloid cells. Together these modular panels provide a robust foundation for accurate end-to-end spatial analyses, even without previous expertise in spatial biology. Abstract
