Spatially interrogating immune cells and associated gene signatures in inflammatory bowel disease 4484

Abstract Description Inflammatory Bowel Disease (IBD) is characterized by chronic intestinal inflammation and includes both Crohn’s disease (CD) and Ulcerative colitis (UC). To effectively treat both diseases, it is important to identify the immune cell profile associated with the type and region of the IBD. The RNAscope™ assay on the BOND RX can enable visualizing the immune cell protein markers with RNA activation markers. Here, we utilized the new protease-free RNAscope™ Multiomic LS assay that can detect up to 6 RNA and protein targets on the same slide with TSA-based amplification strategy that offers a strong signal boost. Here, we demonstrate the use of our pre-conjugated primary antibodies and pre-conjugated secondary antibodies in CD samples. The inflamed tissues demonstrated an overall increased in CD8+ T cells and CD68 positive macrophages compared to uninflamed tissue. Inflammatory gene signatures such as IL-1B and IL-22 among others were also upregulated in CD tissues indicating increased recruitment of immune cells which is associated with the pathophysiology of IBD. The RNAscope Multiomic LS assay allows very specific characterization of immune cell populations by detecting RNA and protein biomarkers crucial in developing targeted next generation therapies for IBD patients. Inflammatory gene signatures such as IL-1B and IL-22 among others were also upregulated in CD tissues indicating increased recruitment of immune cells which is associated with the pathophysiology of IBD.
