Comprehensive evaluation of immune cell biodistribution in whole mice using CryoVizTM Imaging 9013
Abstract Description Monitoring the biodistribution of injected immune cell therapies is critical for evaluating and optimizing their therapeutic effects. Traditional biodistribution analysis often relies on organ harvesting and flow cytometry, which can provide quantitative data but lacks spatial context. The CryoVizTM imaging system creates high-resolution, whole-mouse-field-of-view color anatomical and molecular fluorescence 3-D image datasets by serial sectioning and imaging of the frozen tissue block face, revealing both qualitatively and quantitatively the detailed localization of fluorescent immune cells throughout the whole mouse body. Here, we demonstrate the utility of CryoVizTM imaging to visualize, quantify, and analyze the biodistribution of natural killer (NK) cells, Jurkat T cells, and SUPT1 lymphoblasts in whole mice. In one study, retro-orbitally injected Qtracker 625 (red) labeled NK cells were found in the lungs, liver, spleen, kidneys, bone, and around the eye/lacrimal glands. In a second study with IP and IV injected Jurkat T cells and SUPT1 cells, we found cells in lung, liver, spleen, spine, axillary lymph node, and femur. In a third study with SUPT1 cells, cell biodistribution data suggested that modifying immune cells to overexpress chemokine receptor may stimulate the migration of these cells to the spine – a primary location of whole-body bone marrow. Our results show that CryoVizTM imaging is a powerful tool for evaluating and optimizing immune cell therapies. Topic Categories Vaccines and Immunotherapy (VAC)
