Vital organ specific biomarker signatures in glutenin-induced life-threatening systemic anaphylaxis in a novel mouse model 2479
Abstract Description Failure of vital organs (heart and lungs) is associated with food-induced life-threatening systemic anaphylaxis (LTSA) in humans. We previously reported a novel adjuvant-free mouse model of glutenin-induced (GI) LTSA. Here we tested the hypothesis that a protein microarray analysis method can identify vital organ-specific biomarker signatures in this mouse model. Balb/c mice (n = 10/group) were skin sensitized followed by the induction of GILTSA by systemic challenge. Vital organs were collected from the following groups of mice and used in microarray analysis: vehicle sensitized/vehicle challenged (v/v), vehicle sensitized/protein challenged (v/p), protein sensitized/vehicle challenged (p/v), protein sensitized/protein challenged (p/p). A protein microarray of tissue extracts was conducted by measuring 640 biomarkers. Heat map analysis and fold change in protein expression were used to identify differentially expressed biomarkers. Biomarkers that were increased by three-fold or higher in the vital organs in p/p vs. v/v but not in v/p and p/v groups were identified as GILTSA specific markers as follows: Heart: 26 biomarkers (3- to 468-fold increase); Lungs: 13 biomarkers (3- to 784-fold increase). All the identified GILTSA-specific biomarkers were organ-specific. Here, we report the identification of heart and lung-specific biomarker signatures in a mouse model of GILTSA.
