Targeted silencing of FAM72C impairs cancer cell proliferation 2764

Abstract Description Cancer remains a leading cause of mortality worldwide, with projections indicating 2,001,140 new cases and 611,720 deaths in the United States in 2024. Traditional treatments, such as chemotherapy and radiation, often result in significant side effects. Our prior research, using SNP analysis, identified unusually high expression of the FAM72C gene in prostate cancer cells (LNCaP). To explore its role, we employed RNA silencing, considered a safer method than CRISPR, to reduce FAM72C expression in LNCaP and A-498 renal cell carcinoma cell lines. Fluorescent labeling confirmed successful intracellular delivery of the silencing agents. Subsequent RNA purification, RT-PCR, and qPCR analysis on treated and untreated cancerous and normal cells revealed a significant reduction in FAM72C expression, correlating with decreased cell viability and proliferation in treated cancer cells. These findings suggest that FAM72C plays a crucial role in cancer cell survival and proliferation, highlighting its potential as a target for future gene-specific cancer therapies.
