Antiviral antibody repertoire diversity and dynamics in children and adults 2583

Abstract Description The antibody repertoire, or complete collection of antibodies produced by an individual, reflects an individual’s infection history and particular adaptive immune system. Though it is recognized that individuals exhibit heterogeneous immune responses and both immune memory and capacity to respond to immune stimuli change over the lifespan, cost and complexity have made of population analysis of the complete repertoire prohibitive at scale. We employ VirScan, a high throughput antibody profiling technology, to study the diversity and dynamics of the antiviral antibody repertoire in children and adults. Using large cross-sectional cohorts, we first characterize diversity of the antibody repertoire in adults. We then probe stability of the antiviral antibody repertoire in childhood through adulthood and present a simple algorithm that harnesses antibody repertoire stability to link longitudinal samples from single donors based on their VirScan profiles alone with high sensitivity and specificity. We find that stability and composition of antiviral antibody repertoires are markedly different in children compared to adults and repertoire differences between individuals are much greater than differences between samples taken from a single individual over time. Our results lend insight to population diversity and individual dynamics of the human antiviral antibody repertoire and could inform therapeutic development and healthcare practice.
