Improving the effector function of Vδ2+ γδ T cells through modulating platelet activity 3258

Abstract Description Vδ2+ γδ T cells (Vδ2+) represent a unique subset of T cells distinguished by their expression of the γδ T Cell Receptor (TCR). They account for 1–10% of peripheral blood T lymphocytes and are not restricted by conventional MHC molecules. Platelets are known to interact with T cells, influencing their cytotoxic capabilities. In this study, we investigated the impact of platelets on the cytotoxic function of Vδ2+ T cells, specifically their ability to lyse target cells. Human Vδ2+ T cells were expanded for 18 days with alendronate and subsequently cocultured with platelets at 1:20 and 1:50 ratios for 18 hours. Target cells were then introduced to the Vδ2+ T cell–platelet cocultures at a 1:1 ratio for 20 hours. Flow cytometric analysis revealed that the cytotoxic capacity of Vδ2+ T cells increased by over 20% in target cell killing in a manner that can be reversed by treating the cultures with platelet inhibitors. These findings suggest that platelets may enhance the cytotoxic potential of Vδ2+ T cells, underscoring a promising avenue for developing platelet-mediated immunotherapies.
