Biomarker signatures of intestine and liver in a novel mouse model of glutenin-induced life-threatening systemic anaphylaxis 2474

Abstract Description Wheat allergens such as glutenin can cause life-threatening systemic anaphylaxis (LTSA) in humans. We recently reported a novel adjuvant-free mouse model of glutenin-induced (GI) LTSA. In this study, we tested the hypothesis that GILTSA-specific biomarker signatures of small intestine (SI) and liver (LI) can be identified using a protein microarray analysis. Using the previously published method, LTSA was induced by systemic administration of glutenin in skin sensitized in Balb/c mice. To identify GILTSA-specific signatures, protein array analysis was conducted in four groups of mice (n = 10 per group) as follows: vehicle sensitized/vehicle challenged (v/v), vehicle sensitized/protein challenged (v/p), protein sensitized/vehicle challenged (p/v), protein sensitized/protein challenged (p/p). Tissue protein extracts were used in protein microarray analysis of 640 biomarkers. Differentially expressed biomarkers were identified based on heat map analysis and fold change in protein expression. The markers that were increased by three-fold or higher in v/v vs. p/p but not in v/p or p/v groups were identified as GILTSA-specific signature markers as follows: SI: 22 biomarkers (3- to 35-fold increase); LI: 13 biomarkers (3- to 247-fold increase). All the GILTSA specific biomarkers were organ-specific. We report the identification of GILTSA specific biomarker signatures of SI and LI for the first time.
