Inside Front Cover: Click‐Cyclized Cell Penetrating Peptides Containing Hydrophobic Proline Derivatives for Efficient Intracellular Delivery (Angew. Chem. Int. Ed. 50/2025)
EGFP linked to a click-cyclized cell penetrating peptide that attach itself to the cell membrane and internalize into the cell, while carrying the EGFP as a cargo. The best CPP was selected from a designed library of linear and click cyclized D-peptides having four arginines and a naphthyl group. As indicated by membrane curvature the cell penetration occurs by a dynamin dependent endocytosis mechanism. The cells were healthy at all concentrations investigated indicating low or no toxicity.
