Translation of the optical properties determination method from ex vivo to in vivo for personalization of photodynamic therapy

Personalization of the photodynamic procedure by the assessment of the tissue optical properties should improve the outcome of the therapy. For the determination of the absorption and diffuse scattering coefficients the algorithm based on the Kubelka-Munk theory was proposed. For its development and validation the numerical simulation using Monte-Carlo method and measurements of diffuse reflection (R d ) and transmission (T d ) using the spectrophotometer with integrating sphere Hitachi U-3400 and the set up based on the spectrometer LESA-01-BIOSPEC were performed. The dependencies of Kubelka-Munk (K, S) coefficients from parameters of the theory of radiative transfer were assessed. Using generally accepted relationships between K, S and R d , T d this makes possible the acquisition of the optical properties from spectra measurements. Based on the result of the investigation the suggested methodology for the evaluation of optical properties can be used during the execution of photodynamic therapy.
