The Effect and Mechanism of 5-ALA PDT in Cervical Intraepithelial Neoplasia and Cervical Cancer
Cervical cancer is a major global health issue for women. 5-Aminolevulinic acid-mediated photodynamic therapy (5-ALA PDT) shows promise but has variable efficacy, necessitating in - depth study of its mechanisms and optimization strategies. CCK-8 assay, flow cytometry, Western blot, real-time PCR analysis and siRNA were used to study the effects of 5-ALA PDT on H8 and Hela cells, including anti-proliferative, apoptosis, cell cycle, and gene expression. 5-ALA PDT inhibited the growth of H8 and Hela cells, induced apoptosis, blocked the cell cycle at G0/G1 phase, up-regulated p53, p21, RbAp48, and down - regulated HPV16 oncoproteins E6 and E7. Reduction of RbAp48 weakened the effects of 5-ALA PDT. RbAp48 enhances the sensitivity of H8 cells to 5-ALA PDT through multiple mechanisms, which is crucial for understanding and optimizing 5-ALA PDT in treating cervical cancer.
