Gluten Exorphin <scp>B5</scp> : Synthesis and Characterization of a Food‐Derived Opioid Peptide

ABSTRACT The opioid crisis underscores the urgent need for alternative analgesics that mitigate the risks of tolerance, dependence, and adverse effects associated with conventional opioid drugs. Food‐derived opioid peptides, such as gluten exorphins, represent a promising avenue for safe and effective pain relief. Among these, gluten exorphin B5 (Tyr‐Gly‐Gly‐Trp‐Leu) has garnered attention for its diverse physiological activities, including analgesic, antioxidative, and endocrine‐modulating effects. This study investigates the structural and molecular attributes of gluten exorphin B5, aiming to elucidate the factors contributing to its bioactivity and therapeutic potential. The peptide was synthesized via Fmoc solid‐phase peptide synthesis, purified using RP‐HPLC, and subjected to comprehensive structural characterization. Mass spectrometry confirmed its molecular identity, while NMR spectroscopy provided insights into its three‐dimensional structure. x‐ray diffraction (XRD) analysis revealed its crystallinity, and FTIR spectroscopy elucidated functional group interactions. These advanced analytical techniques facilitated a detailed understanding of the peptide's molecular composition and conformational properties. Results demonstrate that gluten exorphin B5 interacts selectively with opioid receptors, indicating its potential as a food‐derived alternative to traditional opioid analgesics. Despite challenges related to bioavailability and stability, the peptide's structural attributes suggest avenues for optimization. This study highlights gluten exorphin B5's therapeutic promise, paving the way for its development into innovative formulations with broader clinical applications. Future research should focus on enhancing its pharmacokinetics and exploring its long‐term safety and efficacy in vivo.
