The Effect of Eight Weeks of Swimming Training with Narcissus tazetta L Extract on Corticotropin-Releasing Hormone and Mir-25-5p in Rats with Post-Traumatic Stress Disorder

Introduction: Post- traumatic stress disorder (PTSD) is a chronic mental disorder that leads to reduced quality of life and increased economic burden. The present study aimed to review the effect of 8 weeks of swimming training (ST) with narcissus tazetta L (N) flower extract on corticotropin- releasing hormone (CRH) and miR-25-5p in rats with PTSD. Methods: In this experimental study, 24 Wistar rats weighing 280-320 g and aged 14-16 months were subjected to PTSD by 2 hours of bottle immobilization, 15 minutes of forced swimming, and ether anesthesia. Then they were divided into 4 groups including 1) PTSD, 2) PTSD+N, 3) PTSD+ST, and 4) PTSD+ST+N. Also, to examine the effect of PTSD induction, 6 rats were selected as healthy control (HC) group. The ST groups swam for 8 weeks, three sessions per week, with 15-40 minutes of interval ST with 2-minute intervals at 2% of their body weight. The N groups also received 100 mg/kg/day methanolic extract of N orally. The Shapiro- Wilk, independent sample t- test and two-way analysis of variance (ANOA) tests were used for statistical analysis of data (P≤0.05). Results: PTSD induction decreased CRH (HC group: 4.35±0.89 ng/ml vs PTSD group: 1.78±0.46 ng/ml) and increased miR-25-5p (HC group: 1.00±0.38 relative expression vs PTSD group: 2.36±0.4 relative expression) (P=0.001). However, ST and N alone significantly increased CRH and decreased miR-25-5p (P≤0.01), showing no interactive effects (P≥0.05). Conclusion: Although ST and N alone improved CRH and miR-25-5p in PTSD status, nevertheless they did not have interactive effects.
