Multifaceted multivalency with a hub protein
Recent investigations into the interaction between LC8 and GKAP significantly contribute to our understanding of LC8 multivalency. This comprehensive approach addresses four features we find vital for elucidating the mechanisms governing polybivalent complexation behavior: registration control, binding motifs, linker lengths, and flexibility. By deepening our understanding of these characteristics, we move closer to predicting critical aspects of LC8-partner interactions, including whether partners assemble homogeneously or heterogeneously, the degree of flexibility between adjacent binding sites, the strength of binding, and, importantly, the functional consequences of these interactions solely based on the sequence of LC8 binding partners.
