SILENT CONVERSATIONS: JMML'S INTERPLAY WITH T CELL
Juvenile myelomonocytic leukemia (JMML) is an aggressive pediatric myeloproliferative neoplasm driven by constitutive RAS pathway activation. JMML with PTPN11 mutations and DNA hypermethylation is associated with the highest relapse risk following hematopoietic stem cell transplantation (HSCT). While the genetic landscape of JMML is well defined, the mechanisms driving relapse remain poorly understood. We hypothesize that immune escape contributes to disease recurrence. To investigate this, we used high-dimensional mass cytometry to profile the immune landscape of PTPN11-mutant JMML patient spleens.
