Iohexol-Conjugated Polydopamine for Cancer Theranostics

A theranostic platform based on iohexol-functionalized dopamine (IOX-f-DA) and polydopamine (IOX-f-PDA) was developed, offering combined capabilities for precise cancer imaging and targeted therapy. This compound harnesses iohexol’s superior X-ray attenuation for enhanced CT contrast, while polydopamine enables efficient near-infrared photothermal ablation of tumor cells. The biocompatibility of IOX-f-PDA was analyzed through in ovo chorioallantoic membrane (CAM) assays and in vitro evaluation, with negligible toxicity toward normal fibroblasts and selective cytotoxicity upon laser activation in melanoma cells. These findings underscore the system’s ability to integrate real-time imaging precision with localized therapeutic action, marking a significant advance in multimodal cancer theranostics. Importantly, the approach avoids issues typical of small-molecule contrast agents, such as rapid clearance and narrow imaging windows, by leveraging a tailored macromolecular design. Overall, this work establishes a versatile framework for designing dual-modal agents that can improve cancer care’s specificity, safety, and effectiveness and provides a promising direction for clinical development of targeted CT contrast compounds in oncology.
