Zif-8 Loading Gallic Acid For Promoting Bone Defect Repair
To address the challenges of the inadequate control of inflammatory responses and insufficient vascularization which affect the bone defect repair, we engineered a gallic acid (Ga)-integrated hybrid nanocomposite through the strategic incorporation of gold nanoparticles (Au NPs) within zeolitic imidazolate framework-8 (ZIF-8). The nanocomposite was structurally characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), and Fourier-transform infrared spectroscopy (FTIR). In-vitro evaluation of immunomodulation, vascularization, and osteogenesis were performed. The results demonstrated the material effectively suppressed inflammatory responses by inhibiting the NF-κB signaling pathway and downregulating pro-inflammatory mediators. It enhanced endothelial cell-mediated angiogenesis, facilitating early-stage vascularization in bone tissue. Furthermore, it intrinsically promoted osteogenic differentiation. The multifunctional drug delivery system provides a promising technique through spatiotemporal functional coordination to promoting bone defect repair.
